چکیده

Polymorphs of an Active Pharmaceutical Ingredient (API) can have different chemical and physical properties and, thus, a direct impact on the processability of the drug substance and the quality/performance of the drug product, such as stability, dissolution, and bioavailability. Therefore, identification of the crystal phase of an API is of outmost importance. In this work methodologies were developed based on X-ray powder diffraction and IR for the identification of the crystal phase of two APIs (Atorvastatin Calcium and Donepezil HCl) in commercial tablets. INTRODUCTION Polymorphs of an Active Pharmaceutical Ingredient (API) can have different chemical and physical properties and, thus, a direct impact on the processability of the drug substance and the quality/performance of the drug product, such as stability, dissolution, and bioavailability. One polymorph may convert to another during manufacturing and storage, particularly. Therefore, identification of the crystal phase of an API is of outmost importance. Numerous physicochemical measurements and techniques are commonly used to characterize polymorphs, including: melting point, IR, X-ray powder diffraction (XRPD), thermal analysis procedures, Raman spectroscopy, optical microscopy, and NMR. Of them it is mostly Raman spectroscopy and XRPD that have been used for rapid screening of crystal phases. Despite the availability of these techniques polymorph identification in tablets or granules is often very difficult due to: a) the small strength of API in formulation, b) the numerous crystal phases for some APIs and the, as expected, similarity of the respective spectral information and c) the availability in literature of the XRPD patterns only, while information from the other commonly used techniques e.g. IR and Raman spectroscopy are often missing. On the other hand, preparation of polymorphs based on the information mentioned in patents is rarely possible while their acquisition is also difficult and thus the missing spectral information cannot be obtained.

Comparative Study of Patentability of Drug Polymorphism in Iran and the USA

Drug polymorphs are defined as a different arrangement of the same active compounds in crystalline form, which can potentially affect its therapeutic activity by changing the dissolution profile of drug upon administration. Discovery of new polymorphs therefore can provide added value for a previously marketed drug, due to the new advantages provided by the novel polymorph. Therefore, there is ...

Discriminatory Dissolution Test for Tablets Containing a- and b-Thalidomide Polymorphs

Over the years, thalidomide has been prescribed for an increasing number of diseases, including multiple myeloma and erythema nodosum leprosum. In Brazil and other countries, thalidomide is available in tablets, and there is no official dissolution testing available for this dosage form. Considering this, a dissolution method was developed and validated for tablets containing 100 mg of each pol...

A PCA-based chemometrics-assisted ATR-FTIR approach for the classification of polymorphs of cimetidine: application to physical mixtures and tablets.

The identity of the polymorphic form of an active pharmaceutical ingredient is an important parameter that may affect the performance of the drug formulation. This calls for special techniques, able to classify crystal forms or assign the polymorphic identity to a given solid in a mixture. In order to develop a method to determine which of the relevant polymorphs of Cimetidine (CIM) is present ...

Design and Comparative Evaluation of In-vitro Drug Release, Pharmacokinetics and Gamma Scintigraphic Analysis of Controlled Release Tablets Using Novel pH Sensitive Starch and Modified Starchacrylate Graft Copolymer Matrices

The present investigation deals with the development of controlled release tablets of salbutamol sulphate using graft copolymers (St-g-PMMA and Ast-g-PMMA) of starch and acetylated starch. Drug excipient compatibility was spectroscopically analyzed via FT-IR, which confirmed no interaction between drug and other excipients. Formulations were evaluated for physical characteristics like hardness,...

An Investigation into the Effect of Carbopols on the Release of Propranolol HCl from Tablet Matrices

In this study, attempts were made to evaluate the effect of various acrylic acid based Carbopols on the release profile of a beta-adrenoreceptor blocking drug, propranolol HCl, from matrix-type tablets invitro. For this purpose, tablets containing 160 mg of propranolol HCl along with various amounts of Carbopols 934 (C934), 971 (C971), 974 (C974) and Pemulen (Pem) were prepared using the wet gr...
